How to Prepare a Sterile Conical Centrifuge Tube

Before using a conical centrifuge tube, you must ensure that the tube can withstand the pressure required during centrifugation. To prevent mix-ups, it is important to identify the fill level and avoid dislodging the cap during use.
For cell culture applications, sterile conical centrifuge tubes are used. This type of tube has a high-strength, manufacturing process and is designed to provide greater temperature stability. To prepare a conical centrifuge tube, follow the steps outlined below.
Add the correct volume of density gradient media to each sterile conical centrifuge tube. Density gradient media are available in a variety of media including Hanks' Balanced Salt Solution (HBSS) and PBS. Both are saline solutions and contain no calcium or magnesium. However, HBSS is suitable because it is less acidic than PBS.
Next, dilute the blood and the initial separation medium with WDR. After dilution, transfer the sample to the sterile conical centrifuge tube. If the blood is not diluted, the plasma will appear below the density gradient layer and can be difficult to separate. Alternatively, if the entire plasma is diluted, the PBMCs can be separated from the plasma.
Finally, pool the buffy coats and the phage bands into one 50 mL conical centrifuge tube. Label the tube with the PTID and the initials of the technician. Once the sample has been re-centrifuged, remove the phage bands and discard the supernatant.
To maintain the integrity of the sample, it is recommended to follow a protocol for the reagents, such as the Underlay Method. This method is based on the gravitational force between the two layers and creates aggregates. Other methods, such as manual overlay, are also available.
To minimize the possibility of mixing, it is recommended to pipette the initial blood tube on top of the density gradient media. The blood and the density gradient media solution must be pipetted slowly and gently.
If a conical centrifuge tube is not able to handle the pressure of the sample, you can use a re-centrifuge tube. Ensure that the re-centrifuge tube is sterile and can withstand the pressure. It is to keep the re-centrifuge tube at room temperature between uses. Alternatively, the re-centrifuge tube can be stored at low temperature. During the freeze-thaw cycle, the re-centrifuge should be at 2 to 8degC.
When the blood has been diluted, you can add the WDR to it to increase its volume. You can also aliquot the dilution into the sterile conical centrifuge tubes. Add 0.1 mL of the WDR for every 10 mL of blood.
In addition, a PTID should be noted on all tubes of blood received. Using a PTID identifier will help to minimize any cross-contamination. Also, you can mark each PTID with different color markers to avoid confusion.
